Mark Cooper’s Rovers will visit Dean Court in two weeks time.
All ties will take place during the week commencing Monday August 26 with two ties set to be televised.
Michael Flynn’s Newport County – who famously won through to the FA Cup fifth round last season before being beaten by eventual winners Manchester City – have set up another mouth-watering cup tie at Rodney Parade.
League Two County won 4-1 on penalties at Steve Evans’ League One Gillingham after finishing 2-2 over 90 minutes to set up a home second round tie against Premier League West Ham United.
Graham Coughlan’s Bristol Rovers will also host Premier League opposition, Graham Potter’s Brighton & Hove Albion.
League One Rovers eased past League Two Cheltenham 3-0 thanks to two goals by on-loan Sheffield United forward Tyler Smith, who set up strike partner Jonson Clarke-Harris for a third.
Plymouth Argyle will host Championship Reading – managed by Portugal’s Jose Manuel Gomes – following their 2-0 home win over League Two newcomers Orient thanks to goals by Callum McFadzean and Dom Telford.
Neil Warnock’s Cardiff City host newly promoted Championship rivals Luton Town – 3-1 winners over Ipswich Town - while Swansea City are also at home, to League Two Cambridge United following their 3-1 home win over Northampton.
