Clifton, who beat Plymouth Marjon 7-1 on the opening day, were held to a 1-1 round three draw by Lydney following a 9-1 round two win at Isca 2.
Clifton warmed up for next Saturday’s Bristol derby at Firebrands with a 5-0 home win over Newent.
Brands are fourth following a 2-1 win at Isca while Lydney again drew 1-1, with Cheltenham.
Bristol University are third following a 3-1 win at Plymouth, whose scorer was Ruby Westhead.
Promoted Teign, beaten 1-0 by Taunton Vale, and Plymouth seek their first points of the season.
