Reid's new-look young greens' squad has been boosted by the arrival of Hull City midfielder Will Atkinson on a season-long loan - and Reid is hoping his side can be cheered to victory.
The Pilgrims boss said: “The supporters have been brilliant. They know what we are doing, and we need them.
“We need them again today.
“If we can get three points against Rotherham, it will set this season alight.
“The fans at this football club are a big, big influence on the players; the spirit among the players and the fans at this club is second to none.”
Argyle acting chairman Peter Ridsdale, who is addressing what is expected to be a packed meeting of the Argyle Trust at Plymouth Pavilions before the game, agreed.
“We are nothing without our fans, who have been absolutely magnificent since I have been here,” he said.
“We are on the edge of a new era for Argyle, one that will be fully committed to maximising the community nature of the club, and one which will fully engage with supporters at every level. We are all in it together."
