Argyle's 5-1 defeat at Accrington Stanley on Saturday saw the greens slip into the relegation zone following five defeats in a row.
Argyle have only garnered two points in their last eight league games, seeing them tumble from mid-table safety into 21st spot, fourth from bottom of League One.
In an official club statement Argyle "agreed by mutual consent that Derek Adams be relieved of his duties as head of football and first team manager with immediate effect.
"The club would like to place on record its sinceere gartitude to Derek, not only for his hard work and dedication in his management of the team, but also his wider input into the club during four challenging and often exciting seasons.
"Assistant manager Paul Wotton has been similarly relieved of his duties, by mutual consent with immediate effect, and the club also thanks Paul for his contribution and commitment to the Pilgrims' coaching team.
"Kevin Nancekivell will take charge of the (first) team for the vital final match of the Sky Bet League One season at home to Scunthorpe United on Saturday.
"The club will be making no further comment until further notice."
Aberdeen-born former promotion-winning Ross County boss and midfielder Adams, 43, took charge of Argyle in June 2015, leading the greens to promotion from League Two in 2017.
Last season following a memorabe turnaround the greens soared from the foot of the table in December to finish seventh, just outside the play-off places.
Barnstaple-born Nancekivell, 47, is a former Argyle, Tiverton Town and Bideford AFC attacking midfielder. Nancekivell began coaching at Argyle's centre of excellence in 2005 and has subsequently risen up the ranks, becoming first team coach in May last year.
It will be his second spell as Argyle caretaker boss after jointly taking charge of the team with former goalkeeper Romain Larrieu following Carl Fletcher's dismissal in January 2013.
